Nucleic acids and proteins form two of the key classes of functional biomolecules in biology and proteomics. Protein-oligonucleotide conjugates offers us with more possibilities to explore the structure, chemistry and function of proteins. Oligonucleotide conjugation services are convenient so that researchers with little or no conjugation experience can make their own custom protein-oligo conjugate to suit their needs.
Oligonucleotide conjugation service can conjugate various antibodies to oligonucleotides modified by amino groups in the range of 20-60 nucleotides having the best coupling efficiency. Of course, the oligomers with more than 60 bases can also be coupled, and the specific base number needs to be determined after evaluation. The antibodies do not contain carrier molecules such as gelatin or BSA because they interfere with the binding process. Similarly, high concentrations of glycerol (> 25%) are not suitable due to the increased viscosity of these solutions, but small molecule amines such as Tris or sodium azide are acceptable because they are removed during desalination before antibody modification.
Drawing upon years of conjugation expertise, our labeling service offers a protocol for antibody conjugation with oligonucleotide as following:
Stage | Procedure |
Modification of Amino-Oligonucleotide | 1. Resuspend and verify oligoconcentration |
Modification of Antibody | 1.Prepare antibody and verify concentration |
Conjugate Formation and Purification | 1. Conjugate labeledoligo to labeled antibody |
This technology has many practical advantages compared to previous crosslinking methods:
High yielding: routine yields of conjugate are 80% based on starting protein.
High efficiency: >90% of the protein is conjugated.
Stable conjugate: high temperature and wide pH.
High sensitivity and label-specificity enhancement
Suitable for trace protein detection
Mild reaction conditions: no protein denaturation
