Recombinant antibodies are now very important in both therapeutics and diagnostics and offer significant advantages over conventional antibodies because of their high specificity, sensitivity and stability, which may easily be optimised by genetic and chemical modification. There are various recombinant antibodies, including single chain variable fragment (scFv), fragment antigen-binding (Fab) region, single chain antibody fragment (scAb), bivalent scFv (sc(Fv)2), bivalent Fab region (F(ab¡ä)2) and scFv and Fab fusion (Fab-scFv). Single-chain fragment variables (scFv), is a combination of only the variable region of heavy chain (VH) and variable region of light chain (VL) domains. They are mainly constructed by three different methods: scFv generated from synthetic DNA, hybridoma technology or via accession number. A scFv (consisting of VH and V regions) is among the most popular recombinant antibody formats due to its small size, short generation time, capacity to be expressed in Escherichia coli (E. coli) and high antigen affinity and relative structural stability.
